What cell fraction is enriched in the supernatant after cell lysis with cell lysis buffer?

I performed a ChIP assay and I want to know which cell fraction is enriched in the supernatant after cell lysis? is it the cell membrane as it is the largest part of the cell?What cell fraction is enriched in the supernatant after cell lysis with cell lysis buffer?
No.

The cell membranes, nuclei and cytoskeleton and debris, are in the pellet. Light membranes will be in the supernatant together with most proteins and cytosol.



This also depends on the exact method of cell disruption and what buffers were used. You can also get the nuclear proteins in the supernatant.
